Washington State University Collaborator: Lisa G. Neven, USDA-ARS-YARL, Wapato. OBJECTIVES (2005): • Complete development and validation of molecular protocol to discriminate between apple maggot (Rhagoletis pomonella) and snowberry maggot (R. zephyria) captured in traps for accurate monitoring and quarantine actions. • Develop DNA extraction method suitable for flies captured in traps (conservation of specimens far from optimum: the fly dries out and it is immersed in a sticky fluid). • Complete characterization of putative new Grapholita species in roses (rose-hip worm).

A SCAR-based method for rapid identification of four major lepidopterous stored-product pests.
Since Taiwan became a World Trade Organization member in 2002, large quantities of grain have been imported from different countries, and insect pests are frequently intercepted from these imported commodities in quarantine inspection. Because most insects are intercepted as immature forms, morphological identification is problematic; therefore, we developed a DNA identification method based on...

Stored insect pests can seriously depredate stored products causing worldwide economic losses. Pests enter countries traveling with transported goods. Inspection and Quarantine activities are essential to prevent the invasion and spread of pests. Identification of quarantine stored insect pests is an important component of the China’s Inspection and Quarantine procedure, and it is necessary not...
متن کاملTBIS: A Web-Based Expert System for Identification of Tephritid Fruit Flies in China Based on DNA Barcode
Tephritid fruit flies (Diptera: Tephritidae) include serious agricultural insect pests in the world. Besides causing severe damage to fruits and vegetables, this kind of pests could enter countries or regions with international trade easily. Strict trade quarantine measures are imposed in many countries or regions in order to prevent their introduction and spread. Thus accurate and rapid identi...
متن کاملGeneric ionizing radiation quarantine treatments against fruit flies (Diptera: Tephritidae) proposed.
Tephritid fruit flies comprise the most important group of quarantined pests of fresh produce. Most quarantine treatments of fresh agricultural commodities are directed against these pests, and considerable effort in detection, trapping, and population control is expended worldwide to prevent these pests from invading new territories. Ionizing radiation has been studied for 70 yr for its possib...

A diagnostic polymerase chain reaction (PCR) method is presented for differentiating among the North American internal apple-feeding pests codling moth, Cydia pomonella (L.); oriental fruit moth, Grapholita molesta (Busck); lesser appleworm, Grapholita prunivora (Walsh); and cherry fruitworm, Grapholita packardi Zeller. An approximately 470-bp fragment of mitochondrial cytochrome oxidase subuni...
